Limulus ventral photoreceptors were voltage clamped to the resting (dark) potential and stimulated by a 20-ms test flash and a 1-s conditioning flash. At a constant level of adaptation, we measured the response to the test flash given in the dark (control) and the incremental response produced when the test flash occurred within the duration of the conditioning flash. The incremental response is defined as the response to the conditioning and test flashes minus the response to the conditioning flash given alone. When the test flash was presented within 100 ms after the onset of the conditioning flash we observed that: (a) for dim conditioning flashes the incremental response equaled the control response; (b) for intermediate intensity conditioning flashes the incremental response was greater than the control response (we refer to this as enhancement); (c) for high intensity conditioning flashes the incremental response nearly equaled the control response. Using 10-μm diam spots of illumnination, we stimulated two spatially separate regions of one photoreceptor. When the test flash and the conditioning flash were presented to the same region, enhancement was present; but when the flashes were applied to separate regions, enhancement was nearly absent. This result indicates that enhancement is localized to the region of illumination. We discuss mechanisms that may account for enhancement.  相似文献

Many invasive plant species strongly alter ecosystem processes by producing leaf litter that decomposes faster and releases N more quickly than that of native species. However, while most studies of invasive species litter impacts have only considered the decomposition of species in monoculture, forest litter layers typically contain litter from many species. Many litter mixtures decompose in a non‐additive manner, in which the mixture decomposes more quickly (synergistic effect) or more slowly (antagonistic effect) than would be expected based on decomposition of the component species’ litters in isolation. We investigated the potential for non‐additive effects of invasive species’ litter by conducting a one‐year litter bag experiment in which we mixed the litters of four native tree species with each of four invasive species. Litter mixtures frequently lost mass at non‐additive rates, although not at every loading ratio, and the presence, sign, and strength of effects depended on species composition. Non‐additive effects on N loss occurred in more litter combinations, and were almost always antagonistic at 90 days and synergistic at 365 days. Invasive species litter with lower C:N led to more strongly synergistic N loss with time. During the growing season, non‐additive patterns of N loss almost always resulted in increased N release – up to six times greater than would be expected based on single‐species decomposition. Consequently, we suggest that invasive species may further synchronize N release from the litter layer with plant N demand, enhancing any positive litter feedback to invasion. These results highlight the need to consider non‐additive effects of litter mixing in invaded forest communities, and suggest that estimates of invasive species’ impacts on ecosystem processes would be improved by considering these effects.  相似文献

Leaf out time is a widely used indicator of climate change and represents a critical transition point of annual seasonality in most temperate ecosystems. We compared three sources of data to determine the effect of spring temperature on tree leaf out: field observations, remotely sensed satellite data, and experimental warming. All three methods recorded earlier leaf out with warmer spring temperatures. However, leaf out timing was more than twice as sensitive to temperature in the field study (advancing at a rate of 6.1 days/°C), as under experimental warming (2.1 days/°C), with remote sensing intermediate (3.7 days/°C). Researchers need to be aware of the currently unexplained differences among methodologies when using phenological data to parameterize or benchmark models that represent ecosystem processes. The mechanisms behind these discrepancies must be better understood if we are to confidently predict responses of leaf out timing to future climates.  相似文献

Background

Cardiac time intervals have been described as a measure of cardiac performance, where prolongation, shortening and delay of the different time intervals have been evaluated as markers of cardiac dysfunction. A relatively recently developed method with improved ability to measure cardiac events is Tissue Doppler Imaging (TDI), allowing accurate measurement of myocardial movements.

Methods

We propose the state diagram of the heart as a new visualization tool for cardiac time intervals, presenting comparative, normalized data of systolic and diastolic performance, providing a more complete overview of cardiac function. This study aimed to test the feasibility of the state diagram method by presenting examples demonstrating its potential use in the clinical setting and by performing a clinical study, which included a comparison of the state diagram method with established echocardiography methods (E/E' ratio, LVEF and WMSI). The population in the clinical study consisted of seven patients with non ST-elevation myocardial infarction (NSTEMI) and seven control subjects, individually matched according to age and gender. The state diagram of the heart was generated from TDI curves from seven positions in the myocardium, visualizing the inter- and intraventricular function of the heart by displaying the cardiac phases.

Results

The clinical examples demonstrated that the state diagram allows for an intuitive visualization of pathological patterns as ischemia and dyssynchrony. Further, significant differences in percentage duration between the control group and the NSTEMI group were found in eight of the totally twenty phases (10 phases for each ventricle), e.g. in the transition phases (Pre-Ejection and Post-Ejection). These phases were significantly longer (> 2.18%) for the NSTEMI group than for the control group (p < 0.05). No significant differences between the groups were found for the established echocardiography methods.

Conclusion

The test results clearly indicate that the state diagram has potential to be an efficient tool for visualization of cardiac dysfunction and for detection of NSTEMI.  相似文献

Background  

Every cell of the mammalian organism needs iron as trace element in numerous oxido-reductive processes as well as for transport and storage of oxygen. The very versatility of ionic iron makes it a toxic entity which can catalyze the production of radicals that damage vital membranous and macromolecular assemblies in the cell. The mammalian organism maintains therefore a complex regulatory network of iron uptake, excretion and intra-body distribution. Intracellular regulation in different cell types is intertwined with a global hormonal signalling structure. Iron deficiency as well as excess of iron are frequent and serious human disorders. After variable selection, standard inferential procedures for regression parameters may not be uniformly valid; there is no finite-sample size at which a standard test is guaranteed to approximately attain its nominal size. This problem is exacerbated in high-dimensional settings, where variable selection becomes unavoidable. This has prompted a flurry of activity in developing uniformly valid hypothesis tests for a low-dimensional regression parameter (eg, the causal effect of an exposure A on an outcome Y) in high-dimensional models. So far there has been limited focus on model misspecification, although this is inevitable in high-dimensional settings. We propose tests of the null that are uniformly valid under sparsity conditions weaker than those typically invoked in the literature, assuming working models for the exposure and outcome are both correctly specified. When one of the models is misspecified, by amending the procedure for estimating the nuisance parameters, our tests continue to be valid; hence, they are doubly robust. Our proposals are straightforward to implement using existing software for penalized maximum likelihood estimation and do not require sample splitting. We illustrate them in simulations and an analysis of data obtained from the Ghent University intensive care unit.  相似文献

To realistically simulate climate feedbacks from the land surface to the atmosphere, models must replicate the responses of plants to environmental changes. Several processes, operating at various scales, cause the responses of photosynthesis and plant respiration to temperature and CO2 to change over time of exposure to new or changing environmental conditions. Here, we review the latest empirical evidence that short‐term responses of plant carbon exchange rates to temperature and CO2 are modified by plant photosynthetic and respiratory acclimation as well as biogeochemical feedbacks. We assess the frequency with which these responses have been incorporated into vegetation models, and highlight recently designed algorithms that can facilitate their incorporation. Few models currently include representations of the long‐term plant responses that have been recorded by empirical studies, likely because these responses are still poorly understood at scales relevant for models. Studies show that, at a regional scale, simulated carbon flux between the atmosphere and vegetation can dramatically differ between versions of models that do and do not include acclimation. However, the realism of these results is difficult to evaluate, as algorithm development is still in an early stage, and a limited number of data are available. We provide a series of recommendations that suggest how a combination of empirical and modeling studies can produce mechanistic algorithms that will realistically simulate longer term responses within global‐scale models.  相似文献

Background

In sheep dairy production, total lactation performance, and length of lactation of lactation are of economic significance. A more persistent lactation has been associated with improved udder health. An extended lactation is defined by a longer period of milkability. This study is the first investigation to examine the presence of quantitative trait loci (QTL) for extended lactation and lactation persistency in sheep.

Methods

An (Awassi × Merino) × Merino single-sire backcross family with 172 ewes was used to map QTL for lactation persistency and extended lactation traits on a framework map of 189 loci across all autosomes. The Wood model was fitted to data from multiple lactations to estimate parameters of ovine lactation curves, and these estimates were used to derive measures of lactation persistency and extended lactation traits of milk, protein, fat, lactose, useful yield, and somatic cell score. These derived traits were subjected to QTL analyses using maximum likelihood estimation and regression analysis.

Results

Overall, one highly significant (LOD > 3.0), four significant (2.0 < LOD < 3.0) and five suggestive (1.7 < LOD < 2.0) QTL were detected across all traits in common by both mapping methods. One additional suggestive QTL was identified using maximum likelihood estimation, and four suggestive (0.01 < P < 0.05) and two significant (P < 0.01) QTL using the regression approach only. All detected QTL had effect sizes in the range of 0.48 to 0.64 SD, corresponding to QTL heritabilities of 3.1 to 8.9%. The comparison of the detected QTL with results in cattle showed conserved linkage regions. Most of the QTL identified for lactation persistency and extended lactation did not coincide. This suggests that persistency and extended lactation for the same as well as different milk yield and component traits are not controlled by the same genes.

Conclusion

This study identified ten novel QTL for lactation persistency and extended lactation in sheep, but results suggest that lactation persistency and extended lactation do not have a major gene in common. These results provide a basis for further validation in extended families and other breeds as well as targeting regions for genome-wide association mapping using high-density SNP arrays.  相似文献
